The typhoid and paratyphoid bacilli A and B cause greateragglutination when they act simultaneously with their soluble. products. -o-- It is therefore advisable to make the Widal reaction, for the serological diagnosis of typhoid and paratyphoid fever cases, with the r-ulture itself, so as to make use of the agglutinating and precipitating antibodies of the serum on the microbes and their soluble products. The Widal reaction made with cultures in peptonc·water is more specific than when an emulsion of the culture in a solid medium is used, even in the cases when this is prepared in the test tube so as to avoid the loss of the soluble substances. -o-- The serological diagnosis of typhoid and paratyphoid fever cases (Widal reaction) should preferably be made with 24 hrs. old cultures in peptone water. Whenever a culture in a solid medium is used, this should be prepared in the test tube so as to retain the soluble products of the germs. In both cases there is observed a floccuhagglutination.Downloads
